脊椎動物ゲノムにおけるパラロガス領域の総合的解析

研究概要

われわれは、HLA (MHC)領域とパラロガスな領域がヒトゲノムに3箇所存在することを発見し、これらのパラロガス領域とHLA領域の原型が脊椎動物進化の初期におきたと想定される2回の大規模な染色体重複(おそらくはゲノム重複)によって形成された可能性が高いことを示してきた。今日では、本パラロガス群はヒトゲノムで同定された数多くのパラロガス領域のプロトタイプとしての地位を確立しつつある。しかしながら、パラロガス領域を生み出した重複の性質、回数、時期に関しては十分な理解が得られていないのが現状である。そこで、本研究では、系統発生的に2度の重複の直前、直後に位置すると考えられる無顎類(特に、ヌタヌナギ)に焦点を当てて研究を進め、以下の成果を得た。1)哺乳類とほぼ同じゲノムサイズをもつと推定されるヌタウナギのゲノムを3倍カバーするBACライブラリー(平均インサート長約100kb)を作製した。2)上記BACライブラリー(3倍ゲノムカバレッジ分)のスクリーニングを行い、7個のMHC前駆領域をカバーするBACクローンを同定し、その塩基配列を決定した。3)ヌタウナギ白血球由来cDNAライブラリーのランダム・シークエンスを行い、約12,000クローンの5'-end,3'-endの塩基配列を決定した。4)MHCパラロガス群に属するホヤ遺伝子を体系的に同定し、染色体重複後に生じたゲノム構造の変化を解析した。5)免疫グロブリンやT細胞抗原レセプターの祖先と推測される遺伝子群をヌタヌナギで同定し、AAR (ancestral antigen receptor)と命名した。
