遺伝子ネットワーク解明に向けた高機能性人工デコイ核酸の開発

研究概要	細胞内における遺伝子ネットワークを構築している中心的役割を果たすものとして各種の転写因子が挙げられる。これら転写因子は、特異的な配列を認識し遺伝子DNAへ結合することでその遺伝子の発現を抑制したり活性化したりする。そこで、転写因子が結合するDNA配列の構造を模倣し設計・合成した短い核酸断片(デコイ核酸)を細胞へ導入することで、これら転写因子の機能を阻害する、すなわち、転写因子によって制御されている遺伝子の発現パターンを一挙に変化させることが可能となる。これにより、細胞内遺伝子ネットワーク解明が被薬的に進むと期待できる。その実現に向け、我々は天然核酸の糖部コンホメーションを転写因子の結合に適した形に予め固定化した人工核酸(BNA類)の開発研究を展開してきた結果、以下のような興味深い知見を得た。 1)多くの転写因子の結合に適していると考えられるS型糖部コンホメーションを有する新規人工核酸trans-3',4'-BNA類を設計し、その合成に成功した。 2)昨年度までに開発を行ってきた人工核酸3'-amino-3',4'-BNA類の詳細な物性評価を行い、この人工核酸が生体内での安定性にすぐれ、構造的に天然のDNAを完全な形でミミックしたものであることを見いだした。 3)二重らせんの構造を大幅に安定化する人工核酸2',4'-BNAをデコイ分子に導入した。標的となる転写因子は、昨年度までに実績のあるNF-kBとしたが、今年度は新たな配列設計プロトコルの有用性を実証するため、これまでとは異なるストラテジーにてBNAの導入位置や導入個数を決定した。 4)上記NF-kBを標的とした評価系において、人工核酸を導入したデコイ核酸やアンチセンス分子を用いることで、NF-kBの機能阻害に伴う細胞のアポトーシス死を確認し、そのアポトーシスに至る経路に関与する遺伝子群を確認することに成功した。
