シロイヌナズナ完全長cDNAマイクロアレイを用いた遺伝子発現解析

研究概要	本研究では、モデル高等植物のシロイヌナズナを用い、シロイヌナズナDNAマイクロアレイを用いた解析により、種々のストレス、ホルモンに応答する遺伝子等を単離、同定する。同定された遺伝子に関してプロモーター解析を行う。これまでに単離した約7000個のシロイヌナズナ完全長cDNAクローンを用いて完全長cDNAマイクロアレイを作製し、乾燥,低温、塩などの種々のストレスまたはABAなどの種々のホルモン処理、種々の変異体やトランスジェニック植物など200種類以上の異なる条件下での発現プロファイル解析を行った。論文発表したものの幾つかに関しては発現データを公開した。アレイ解析により乾燥2時間後の再吸水処理により応答する遺伝子を152個同定した。同定された誘導性遺伝子に関して再吸水処理過程での発現パターンについて解析し、再吸水処理直後には、"細胞の乾燥ストレス状態からの解放"に関わる遺伝子が機能し、やがて、後期になって、"光合成や植物生長などの再開"に関わる遺伝子が機能する事を明らかにした。再吸水処理誘導性遺伝子の中には、乾燥ストレスにより発現誘導されるもの、発現が抑制されるもの、発現に変化がないものの3つのグループが存在することを明らかにした。再吸水処理誘導性遺伝子と無処理植物体を単に水に移すという処理で発現誘導される遺伝子とをベン図解析して、再吸水処理誘導性遺伝子の中には1)単に水に移すという処理でも発現誘導される遺伝子(細胞内の浸透圧変化の調節に関与するもの)と、2)単に水に移すという処理では発現誘導されない遺伝子(乾燥ストレスにより生じたダメージの修復に関与するもの)、の2つのグループが存在することを明らかにした。さらに再吸水処理誘導性遺伝子のプロモーター解析の結果、再吸水処理による発現誘導に関与する新規なシス配列の存在が示唆された。
