網膜マイクロアレイと候補SNPを用いた糖尿病網膜症の遺伝素因の同定

研究概要	SNPを用いた2型糖尿病との関連解析において最も重要なことは均一集団を設定することである。そこで申請者は、体外から詳細に「病気の質」が目で観察できる増殖網膜症に注目した。候補遺伝子について検出感度の高いSNPマーカーを開発し、病型との関連解析を実施した。候補遺伝子の選別に対しては、2種類のアプローチを試みた。 1)HIF候補遺伝子の解析血管新生において血管内皮増殖因子VEGFが重要な働きを担う。そこで、VEGF遺伝子の転写因子Hypoxia inducible factor (HIF)のコード遺伝子解析を前年度に開始した。13個のSNPを求めて、168人の増殖網膜症を有する症例と185人の非糖尿病者の間で関連解析を行ったところ、3'端のSNP-13の出現頻度に有意差を認めた(P=0.0171)。連鎖不平衡を考慮してSNPハプロタイプを構築し、300サンプル数を用いて成績を確認した。このSNPは近隣のcSNP(ミスセンス変異)と有意の連鎖不平衡にあったので、同SNPが原因サイトである可能性が示唆された。現在、PCR mutagenesis法により変異導入を行ない、変異蛋白の機能解析を行なっている。 2)網膜ESTマイクロアレイの作成 17,184個の3'ESTを用いたクラスタリング解析により、約半数のESTが未知遺伝子由来であることを既に明らかにしている。この内からハウスキーパーを除去し、先ず既知遺伝子を中心としたアレイを作成した。同アレイを用いてGKラットの糖尿病発症前後の網膜組織について解析を行なった。現在、発現変化遺伝子の区分を行なっている。
