1型糖尿病遺伝子の機能解析に基づくテーラーメード医療の構築

研究概要	1型糖尿病のモデルNODマウスにおいて、疾患感受性に最も強く関与するMHCの領域のIdd1に関して第2のコンポーネント(Idd16)をコンジェニックマッピングによって限局するとともに、領域内の強力な候補遺伝子であるクラスI MHCのK遺伝子の全exon,exon-intron junctionならびに5'上流領域(-700bp)の塩基配列を決定した。その結果、Idd16に抵抗性アリルを有するCTSマウスとIdd16に感受性アリルを有するNODマウスとの間に10個の一塩基置換(SNP)を見出した。その内訳は、5個がプロモーター領域、1個がエクソン2、4個がエクソン4に存在した。ブロモーター領域の多型は既知の転写因子結合部位に影響を与えるものではなかったが、エクソン4の3つのSNPはTyr^<191>(NOD)→His^<191>(CTS)、Asp^<198>(NOD)→Lys^<198>(CTS)という2つのアミノ酸置換をもたらし、K分子のα3ドメインの構造に変化を与えることが示唆された。NODはK^dタイプと同一の塩基配列であったが、CTSおよびNOD.CTS-H-2コンジェニックマウスではK^dタイプの分子がこの2アミノ酸のみK^kタイプに置換した全く新しい分子であることが示された。この塩基置換がアミノ酸置換を介してK分子の構造に変化を与えることを検証する自的で、NOD、CTSおよびNOD.CTS-H-2コンジェニックマウスのK分子をモノクローナル抗体を用いたFACS解析により比較したところ、NODマウスがK^dに対する抗体とのみ反応したのに対して、CTSおよびNOD.CTS-H-2コンジェニックマウスではK^dに加えてK^kに対する抗体とも反応を示し、塩基配列レベルの変化が分子レベルでも反映されていることが示された。以上の結果より、Idd16の原因としてクラスIのK遺伝子の多型が関与している可能性が示された。ヒト対応領域を検索した結果、ヒトでもクラスI領域に病態に影響を与える遺伝子が存在する可能性が示された。
