放線菌ストレプトミセス・グリセウスのA-ファクター制御カスケードの網羅的解明

研究概要	放線菌Streptomyces griseusの抗生物質生産と形態分化は、自身の生産する化学調節物質A-ファクターによりスィッチ・オンされる。本研究は、A-ファクター制御カスケード内の遺伝子を網羅的に解明することであり、本年度の成果は以下のようにまとめられる。 1.S.griseusのゲノム配列決定に着手し、これまで約40,000サンプルにつき塩基配列決定を行った。アセンブリーと平行して、コスミドクローンの末端配列の解析を行っており、来年度中には全配列決定、遺伝子のアノテーシヨンを終了できる見込みである。 2.A-ファクター制御下にある黄色色素(グリキサゾン)合成遺伝子群につき、A-ファクターシグナルを受容するシスエレメント領域を同定した。また、6個の転写単位を同定し、これら全てがA-ファクターおよび培地中のリン酸濃度に応答することを見出した。 3.A-ファクターカスケード内の正の転写因子AdpAの標的遺伝子として新たにトリプシン型プロテアーゼ2種キモトリプシン型プロテアーゼ3種を見出し、A-ファクターによる転写制御様式、AdpAの結合塩基領域および遺伝子破壊による形態分化に及ぼす影響を検討した。 4.A-ファクター制御カスケードとcross-talkを行うたんぱく質リン酸化系であるAfsK/AfsRにつき、TOF-MSを用いてリン酸化残基を決定した。現在同定されたアミノ酸残基に部位特異的変異を行い、最終確認を続行中である。
