植物SRY遺伝子:ヒロハノマンテマのY染色体特異的ゲノム構造と雄性決定遺伝子

研究概要	本研究の目的は、ナデシコ科の雌雄異株植物、ヒロハノマンテマSilene latifoliaのY染色体にある雄性決定領域とそこにコードされた雄性決定遺伝子(植物SRY遺伝子)を同定することにある。こうした研究を通じて、1)植物の巨大ゲノムの一部(本研究課題ではY染色体)のみを研究対象として取り扱う手法を確立し、2)高等植物にも拘わらず性染色体が発達した意味を明らかにする。具体的な研究目標は、1)BACライブラリーを拡充し、2)Y染色体上腕末端にある繰返し配列やSTSマーカーを指標にY染色体特異的なBACクローンのコンティグを構築する。3)コンティグを精査し繰返し配列やSTSマーカー近傍からY染色体特異的な遺伝子を見出す。4)ヒロハノマンテマあるいはアラビドプシスを用いた性転換系を確立し、新に単離されたY染色体特異的な遺伝子の性転換能を検証できるシステムを構築する。平成15年度の具体的な研究実施計画に関しては、高等植物から無傷のDNAを大量に単離する方法を確立し、Y染色体上腕末端に特異的な繰返し配列を指標に、BACクローンのコンティグを構築することを研究の中心とした。具体的には、1)BACライブラリーの拡充:13年度以降細胞核の単離法に改良を加え、BACクローンの獲得数を飛躍的に増大させた。現在のBACライブラリーは、クローン数32,642、インサートサイズ50〜200kbで、ゲノムのカバー率は約80%となっている。2)Y染色体特異的BACクローンの単離:Y染色体特異的STSを用いた4D-PCR高速スクリーニングで上記のBACライブラリーから7つのY染色体特異的BACクローンを単離した。3)BACクローンのシーケンスとY染色体特異的遺伝子の検出:FISH法によってY染色体特異的配列の蓄積が最も著しいと推定された127kのインサートをもつMS2-9d12Fの前塩基配列をショットガンシーケンスし、47個のORFを同定した。
