脳特異的新規膜貫通蛋白質ファミリーの神経発生における役割

研究概要	神経突起は神経細胞を特徴づける細胞構造のひとつであり、神経細胞サブタイプによって、特有な数、長さ、分枝パターンを示す。神経突起の伸展、極性を調節する可能性のある新規蛋白質として、Slitrkファミリーを同定し、解析に着手した。 Slitrkファミリーは膜貫通蛋白質であり、マウスでは少なくとも6種類の関連遺伝子が存在していた。C末端側にはニューロトロフィンレセプターと相同性のある領域に保存されたチロシン残基が存在しており、リン酸化チロシンを介した細胞内情報伝達に関わる可能性が高いものと考えられた。Slitrk1-6を培養神経細胞で発現させたところ、あるものは軸索に似た一本の突起を誘導し、別のものは神経突起伸展を著しく阻害した。トランスジェニックマウスを作製し、Zic1プロモーターの制御下に脊髄の背側の介在神経細胞に発現させたところ、交連線維を含めた神経突起の形成に異常が認められた。 Slitrkファミリーの発現は神経細胞の成熟段階、あるいは神経組織の領域に依存する傾向があり、大脳皮質、間脳、脊髄運動ニューロンなどで特徴的な発現を示した。特に大脳皮質ではSlitrk1-6が異なったパターンで層状に発現し、間脳では背側視床(prosomere2の領域)でSlitrk6が限局的に発現していた。また、ヒトSlitrk1-6はさまざまな脳腫瘍で、腫瘍のタイプにより独特なパターンで発現していることが明らかになってきた。また、特にSlitrk6のみは発現プロファイルが他のメンバーと異なり、中枢神経外の組織でも、頭頚部の外感覚器、消化管などの特定の領域での発現が認められた。以上の結果から、Slitrkファミリー遺伝子の発現制御機構は神経組織の分化・成熟・領域化の点で興味深いものと考えられた。現在、その分子機能に関する解析をさらに続けている。
