微生物感染における炎症と免疫反応のケモカインによる分子制御

研究概要	血中樹状前駆細胞ホーミング機序の解析正常及び炎症時のマウスの全身リンパ節・高円柱内皮細静脈(HEV)の詳細な免疫3重組織染色により、血中,pDC前駆体がHEVを介して血中からリンパ節に直接遊走し得る細胞群である事が強く示唆される結果を予備実験で得ていた。引き続き、血中mDCとpDC前駆体のホーミング分子をセレクチン、ケモカイン、インテグリンに着目し、FACSならびにPCRで発現パターンを解析した。pDC前駆体に関してはL-セレクチンとCXCR3がより高度に発現していたため、その機能を調べるべくまず凍結切片上でのHEV結合試験を行った。その結果、樹状細胞はリンパ球と異なり、正常リンパ節のHEVにはほとんど結合しないことが判明した。炎症時のリンパ節のHEVには、所属、非所属リンパ節を問わず結合する細胞群が認められ、これらがbDC前駆体のみであり、かつCXCR3のリガンドであるCXCL9/Migの中和抗体前投与によりHEVとの結合がきれいに阻害された。一方、L-セレクチンの中和抗体による阻害実験ではこのような阻害を認めず、逆にE-セレクチンの中和抗体でHEVとpDC前駆体の結合が阻害された。リンパ節HEVの免疫染色では、正常時にはほとんど認めなかったCXCL9とE-セレクチンが、P.acnes感染時には一過性ながら極めて強く発現するようになることを確認できた。以上の結果をもとに、短時間移入実験での阻害実験を試みた。同様に、pDC前駆体のみが炎症マウスの全身リンパ節に侵入することが明らかになった。このHEVを介した短時間のリンパ節遊走はCXCL9とE-セレクチンの共阻害によりほぼ完全に抑制された。一方、mDCは全身の末梢臓器に遊走し得たが、この短時間ではリンパ節への直接遊走は認められなかった。
