HIV特異的分泌型IgA誘導粘膜ワクチン開発へ向けての基礎研究

研究概要	粘膜免疫システムを駆使してHIVに対する第一線のバリアをその侵入門戸である生殖器粘膜に構築するには、経口・経鼻ワクチンが有効であり、その鍵を握っているのが粘膜免疫増強効果のある粘膜アジュバントや新型粘膜ワクチンデリバリーシステムの開発である。そこで第一世代としてコレラ毒素(CT)を無毒化した変異型(mCT)を開発した。さらに第二世代としてB.choshinensis発現ベクターを使ってmCTサブユニットと(mCT-A)と病原性大腸菌由来易熱性毒素のLT-Bサブユニット遺伝子(pNCMO2-LTB-mCTA)を発現しているキメラ分子(mCT-A/LT-B)を作成し、HIVをはじめとする各種ワクチン抗原特異的粘膜免疫増強効果について検討してきた。キメラ型ではCT-Bに比較してLT-Bが宿主細胞の発現する多数のリガンド分子(GM1,GM2,asialoGM1)に結合できる特徴をいかして、粘膜上皮細胞へのワクチン抗原などの送達効果の増強が期待できる。同キメラ型アジュバントの無毒性については腸管結紮ループ試験、cAMP誘導性試験などを併用して確認してきた。さらにキメラ型にもワクチン抗原に対して粘膜アジュバント効果がある事を確認した。次に第一世代のmCTに関してはマウスでの実験を終了し、将来のヒトへの応用を踏まえて霊長類での検討を開始した。HIVgp160をmCTと混合し、rhesus macaqueに経鼻投与すると、抗原特異的IgAと抗体が誘導出来る事が明らかになった。つまり、ヒトにより近い霊長類でもmCTを粘膜アジュバントとしてHIVワクチン候補抗原と一緒に経鼻免疫すると抗原特異的な粘膜系IgAと全身系IgG両方が誘導できる。今後は、HIV抗原特異的分泌型IgA(例:生殖器や腸管分泌液、唾液)や血清IgG抗体の中和効果を検討するさらにHIV特異的CTLの誘導に関しては生殖器や腸管上皮細胞間リンパ球(IEL)を分離解析する。
