研究概要

親株(Con1株)C型肝炎ウイルス(HCV)サブジェノミックレプリコンをもとに、リバースジェネティクスにより、NS5Aの種々の変異を持つHCVレプリコンを作製し、Huh-7細胞に導入してレプリコン複製細胞株を得た。導入した変異は第37位アミノ酸Phe->Leu変異(F37L)、Phe->Asn変異(F37N)、その他種々のISDR点突然変異等である。まず、F37Nを有するHCVレプリコン複製細胞株は全く得られず、F37NはHCV遺伝子複製にとって致死的変異であることがわかった。種々のNS5Aと2'-5'オリゴアデニル酸合成酵素(2-5AS)を発現する細胞における両蛋白の結合実験より、F37N変異NS5Aは2-5ASとの結合が弱いこと、また、脳心筋炎ウイルスのレスキュー実験より、F37N変異NS5Aはインターフェロン(IFN)抗ウイルス作用の抑制活性が弱いことを明らかにした。一方、F37L変異NS5Aは2-5ASとの結合が増加し、より強いIFN抑制作用を示した。F37L変異HCVレプリコンのIFN抑制作用は親株レプリコンと同程度であった。ISDR変異については、IFN応答性に関する臨床成績から重要と考えられる種々の点突然変異(T244A, I255V, D248G,3点〜8点変異など)を導入したHCVレプリコンを作製した。それらの変異レプリコンは親株と同程度によく複製した。これらのISDR点突然変異のうち、8点変異を除いて、IFN抗ウイルス効果の抑制は親株と同程度であった。現在、8点変異及び他の部位のISDR変異を有するレプリコンを作製し、IFN抗ウイルス効果に及ぼす影響についてさらに詳細に解析を進めている。
さらに、Akt/Protein kinase B(PKB)に及ぼすHCV全蛋白の影響を調べる目的で、全長HCV遺伝子発現レプリコン複製細胞を作製している。Akt/PKB酵素活性の測定法を確立し、コア蛋白がPDGFによるArt/PKBの活性化を増強することを明らかにした。今後、全長HCVレプリコン複製細胞株における。Akt/PKB活性について詳細に検討する予定である。
