CCR5を標的とした新規の抗HIV剤の基礎的研究と開発

研究概要	本研究では既存の薬剤と異なる作用点を持つ抗HIV剤の開発を目的にケモカインレセプター(CCR5)阻害剤の開発・研究を展開した。新規のCCR5阻害剤:AK602/ON04128は我々が見いだしたプロトタイプの化合物:E913(Maeda, Miyakawa & Mitsuya, JBC.276:35194-35200,2001)より強力な抗HIV活性と優れた薬物動態を示した。更に複数のAIDS治療薬に対して高い耐性を獲得した変異株を含めたR5 HIVの感染・増殖を試験管内で強力に抑制し、動物モデル(NOD-SCIDマウス)の系でも強力な抗ウイルス活性を発揮、HIVウイルス血症を約2log抑制することを示した。更に放射線ラベル化(^3H)化合物を用いた検討でAK602は強力なCCR5に対する結合親和性K_D値:2.9nM)を持ち、さらにレコンビナントgp120とCCRとの結合を強力に阻害した。その一方でAK602はケモカイン、特にRANTESの生理的作用にはわずかしか影響を与えない結合様式をとっていることが分かった。更にCCR5阻害剤と種々の組み換え型CCR5を用いてAK602が既存のCCR5阻害剤の結合部位と異なる部位に結合、その中でもextracellular loop2の両端に位置するアミノ酸(G163とK191)が結合に重要であることを示した。 AK602/ONO4128は米国での健常者での初期安全性試験(Phase1)が終了、現在、臨床第2相試験が行われている。本研究の意義としては、新規のCCR5阻害剤:AK602のin vitro/in vivoの評価を進め、臨床試験段階まで進めたことと共に、CCR5阻害剤とケモカインの相互作用、CCR5との結合様式といった基礎的な知見も多く得られたことが挙げられる。
