サルを用いたHAARTと免疫療法の前臨床動物モデルの確立

研究概要

今年度我々は、昨年度から引き続きHIVとSIVのキメラウイルスであるSHIV_<89.6P>を感染させたマカクサル(慢性感染系)を用いて、経口的に臨床で行われている薬剤の組み合わせでの動物モデルの構築が可能であるかどうかを検討した。SHIV_<89.6P>を用いたin vitroでの薬剤感受性実験を行い、HAARTに用いる薬剤を3種類選択し(AZT+3TC+LPV/r)、これらを、SHIV_<89.6P>を接種したマカクサルに、4週間経口的に投与したところ、ヒトで見られるのと同様に、血中のウイルス量とPBMC中のプロウイルス量の著明な低下がみられ、HAARTの中断後に急激なリバウンドが見られた。また、現在HIV感染患者において研究が進んでいる、治療の有効性の評価の指標(CD4数、RNAウイルスコピー数、proviral DNAコピー数、T細胞のターンオーバーなど)が、人の場合と同様に、キメラウイルス感染サルにもあてはまることが確認でき、報告した(Yoshimura K.et al.,J Virol Methods,2003)。このことから、より実際の治療に近い薬剤の組み合わせと投与方法を行うサルモデルの構築が可能であることが証明できた。今年度以降は、再度この感染系を用いて、多剤併用療法を行いつつワクチンを接種したり、HIV化感染症例で使用予定の治療用単クローン抗体とHAARTを組み合わせて投与するための前臨床テスト等を行う予定にしている。
