研究概要	腸管出血性大腸菌(EHEC主にO157:H7)感染症は、小児、老人などの免疫力の低下したヒトに多く発症し、死亡例が後を絶たない。本感染症は、発症菌量が数十個とされ極少量で重篤な症状を起こし、無症状キャリアー成人からの家族内感染が多く報告され、感染経路の特定が困難な場合が多い。EHECによる感染症はいまや日本中で集団感染、弧発例が見られその感染経路も多様化している。本菌のキャリアーは家畜、特にウシとされている。いまや世界中に蔓延している人畜共通感染症である本症を撲滅するには、ヒトおよび飼育されているウシからの本菌の排除が必要である。ところが、ウシが飼育されている国は世界中に及び、また、ワクチンなどの投与が経済的に不可能な地域が多く含まれる。そこで、安価に栽培でき、ヒトおよび家畜に経口摂取可能な植物ワクチンが有用であると考える。 EHEC(O157:H7 Sakai株)由来のタイプIII分泌機構に関連するEspA,EspB,intimin、及びベロ毒素Bサブユニットをコードする遺伝子を用いて12種類の発現ベクターを構築し、日本固有のマメ科植物ミヤコグサに導入した。約600本のシュートの中からPCRによるスクリーニングで298個体の形質転換体を選択した(T_0)。遺伝子導入が確認された8種類の再生個体を育成し、221個体から各々のT_1種子を採取、ベクターの種類ごとに100個体(計800個体)の種子を播種した。一般的に植物においては複数コピーの外来遺伝子が導入されている場合RNAiによるpost transcriptional gene silencingが起こる。そこでT_1個体の一部を用いてSouthern blotなどを行ない、単一コピーの遺伝子が挿入されている13個体を選別した。これらの13個体中T_17個体を育成した結果、T_2計89個体を得た。今後、小動物においてワクチン効果を解析していく。
