研究概要	Hodgikin細胞のの多核状態は免疫グロブリン遺伝子が再構成していて、すべてV領域の突然変異が著しく亢進し、それらがすべて著しい変異のために終止コドンが入り、抗体分子ができない。これらは胚中心B細胞における体細胞遺伝子突然変異誘導のメカニズムに何らかの破綻をきたし、その結果特異なガン化状態を引き起こしたことを示唆している。研究計画ではタイプ2RNAプライマーゼGANPのDNA複製とその修復に関わる機能とガン化のメカニズムの関連を明らかにする。ヒトリンパ腫細胞ことにHodgikin細胞ではリン酸化型とGANPが著しく発現上昇し、腫瘍細胞の特定、診断に有効であることが示された。この分子の過剰発現B細胞株を作成すると、その細胞では著しく、免疫グロブリン遺伝子のV領域に体細胞突然変異が誘導された。このことからGANP分子の発現が抗体分子の親和性の亢進のためのDNA変異導入に機能する可能性を示した。さらに、この遺伝子を過剰発現したトランスジェニックマウスを作成したところ、このマウスのほぼ過半数以上に50週令を経過するとリンパ腫様の腫瘍が発症した。胚中心で発現が上昇するGANP分子は免疫応答で抗原の刺激とCD40からの刺激を受けると活性化されるDNA複製関連分子と考えられる。本研究ではこの分子の発現異常や、構造異常が実際にリンパ細胞の腫瘍化に関連しているかを解析した。これまでのin vitroおよび、遺伝子導入トランスジェニックマウスにおける生体の解析でGANP分子の異常発現が効率にリンパ腫細胞の発症と関連していることを初めて明らかにした。この結果は、臨床病理検査におけるヒトのリンパ腫の解析結果とほぼ一致することから、これまで以上に、診断、治療を明確にすることが可能となり、また疾患の予防や治療の戦略に有効な情報を提供するものと考えている。
