スフィンゴ糖脂質によるraftの構造調節と増殖・分化シグナルの制御機構

研究概要	細胞膜上のミクロドメインであるraftはシグナル伝達の場として重要である。酸性スフィンゴ糖脂質(ガングリオシド)GM1はそのマーカーであるのみならず、raftの性状に影響を与える重要な分子と考えられる。一方、神経外胚葉由来の癌細胞にはガングリオシドGD3、GD2の発現の増強が認められ、癌の悪性形質に関与している可能性がある。これらに関連して本研究では次のことが明らかになった。GM1高発現ラットPC12細胞ではNGF刺激による分化が抑制されたが、これは細胞膜上のGM1によりNGF受容体(R)がraftに局在できず、その結果、NGFによるNGFRの2量体化の形成が阻害されシグナル伝達が起こらないものと推察された。一方、EGF刺激では、低血清下においてGM1高発現細胞の細胞増殖の亢進とEGFR及びMAPKのリン酸化の維持が認められた。この場合はEGFRとraft局在は無関係で、GM1によるEGFRの修飾が考えられた。また、ガングリオシドGD3を高発現させたヒトメラノーマ細胞では、細胞の増殖と浸潤能の亢進が認められた。このGD3発現細胞はFCS刺激によるMAPK、Akt、FAK、p130Cas及びpaxillinのリン酸化の増強が認めらた。本システムにおいて、これらの分子の細胞増殖・浸潤に対しての関与の有無を明らかにするためにRNAi法により各分子をノックダウンを行った。現在のところp130Cas及びpaxillinがGP3高発現細胞において、その浸潤性に関与していることが明らかとなった。今後GD3とこれらシグナル分子との関連を詳細に検討し、スフィンゴ糖脂質の癌細胞の悪性形質における役割について検討を進める。
