破骨・造骨カップリング制御による骨破壊腫瘍の制御

研究概要

本研究では、破骨細胞・骨芽細胞の相互作用を分子レベルで解析し、骨破壊性腫瘍疾患において、腫瘍の骨への浸潤・破壊の制御・抑止をはかることをめざした。
DNAサブトラクション法による破骨細胞特異的分子の検索を行い、他の血液細胞系列の細胞や組織での発現解析から破骨細胞に特異的な分子を絞り込み、解析を進めた。
1)SU166:膜7回貫通型蛋白で、そのコーディング領域に蛍光色素(GFP)遺伝子を挿入した遺伝子破壊マウスを作製した。予測通り、破骨細胞や脳ミクログリアなどに、GFPの発現がみられた。また、予想外なことに、腎臓や卵巣の一部にも発現がみられたので、現在その詳細を検討している。以後、ホモ欠損マウスにおいて、骨異常がある否かを中心に解析を進める。
2)SU180:heat shock proteinで、アポトーシスに関連があると考えられ、現在siRNAを用いて検討を進めている。
このように、本研究において、破骨細胞にする遺伝子を多種クローニングすることができ、今後、研究を展開するうえでのseedsをつくることができた。
また、骨髄造血研究の進展により、造血幹細胞と骨芽細胞の接着が明瞭となった。この研究および1)の研究により、骨梁部分と骨幹部分では、それぞれ骨芽細胞や破骨細胞の形状・機能が異なるかもしれないことが示唆された。これは、転移性骨腫瘍の骨破壊を克服するうえでも極めて重要な視点を与えるものと考える。
