増殖因子REGを分子標的とした新規抗がん剤リード化合物の探索

研究概要

1.REG(Regenerating Gene)遺伝子及びその関連遺伝子(REG遺伝子ファミリー)の転写調節領域を分離し、ルシフェラーゼ遺伝子の上流に挿入したレポータープラスミドの作製を行った。
(1)この過程で、従来REG遺伝子ファミリーは4つの機能遺伝子(REG Iα,REG Iβ,HIP/PAR,REG IV)と1つの偽遺伝子(REG-related sequence)から構成されていると理解されてきたが、新しいREG関連機能遺伝子を見出した(論文投稿中)。
(2)上述の新規遺伝子を含め全ての遺伝子の上流約1kbpをルシフェラーゼ遺伝子の上流に挿入したレポータープラスミドを構築した。
2.大腸・直腸がんや胃がんなど種々のがん細胞でREG遺伝子ファミリーの遺伝子発現をRT-PCRで検討した。大腸・直腸がんではSW948,SW1417,COLO205,HCT15、胃がんではKATO III,MKN28,AGSなど多くの細胞でREG遺伝子ファミリーの発現が認められた。
3.SW948細胞にREG遺伝子ファミリーのレポーター遺伝子を導入し、ルシフェラーゼアッセイを行ったところ、ルシフェラーゼ活性はRT-PCRで確認された遺伝子発現量に対応していたので、REG遺伝子ファミリーの発現は、転写レベルで調節されていることが明らかになった。
4.REG遺伝子ファミリーのうち、ヒト大腸がん患者の生命予後と強い関連を示すのはREG Iαの発現である。そこで、REG Iαのレポーター遺伝子を導入したSW948細胞の培養培地に種々の化学物質を添加し、レポーターアッセイによりREG Iα遺伝子発現を抑制する化合物のスクリーニングを行った。この結果、数種類のREG Iα遺伝子発現低下を引き起こす化合物質を見出した。
