ファージライブラリーを用いたIL-6阻害ヒト抗体および低分子癌抑制分子の開発

研究概要

サイトカインは腫瘍細胞の増殖因子であり、その機能阻害による癌の治療が期待される。抗TNFα抗体や抗IL-6受容体抗体を用いた治療はこのような知見から非常に有効な治療手段といえる。しかし、これらの抗体は高分子であるため投与方法に制限が生じる。そこで、我々はより利便性の高い阻害剤の開発を目指している。
まず、ヒト抗体を用いたサイトカイン阻害剤としてsingle chain fragment variant(scFv:H鎖とL鎖の可変部のみの遺伝子を結合させた異型抗体)を導入した抗体ファージライブラリーを用いてIL-6に結合するscFvの発現クローンを得る方法を確立した。これらのscFvからIL-6依存性細胞増殖抑制能を持つクローンを選別した。
つぎに無差別配列した7アミノ酸より成るペプチドを表現したM13ファージライブラリーを用いてリコンビナントIL-6をコートしたプレートにまき、IL-6結合ファージクローンを得た。IL-6特異結合クローンを選択するため、IL-6、ヒトIg、ヒトアルブミン、BSA、またはゼラチンをコートしたディッシュを用いて、得られた18クローンを反応させ、ビオチン化抗M13抗体にて反応させた。その結果、IL-6に特異的に結合するクローンを3-5種得、さらに数を増やすため2-3回同法を行った。その後IL-6依存増殖する細胞の増殖阻害を行い数種のIL-6増殖抑制クローンを得た。
今後IL-6の特異性、あるいはペプチドによる増殖抑制を検討する。
