バイオ・サーファクタントを用いた正電荷リポソームによる遺伝子治療

研究概要	アデノシンデアミナーゼ欠損症(ADA欠損症)の患者に対して試みられた遺伝子治療の研究は、新しい医療の幕開けを告げるものであった。それ以来、遺伝子治療は遺伝病の根元的治療法になるだけでなく、癌やエイズの治療にも有効であると期待されており、今後の重要な医療技術の一つになることは間違いないであろう。しかし、遺伝子治療を受ける患者数の増加とともに、病原性ウイルスをベクターとして用いた場合の不慮事故の報告数が増加の一途をたどっている。この問題を解決するため、研究代表者らは正電荷コレステロール誘導体を素材とした安全性の高い非ウイルスベクター(正電荷リポソーム)の開発に取り組んできた。代表者らの新規ベクターは高い安全性はもちろんのこと、遺伝子導入効率においても既存の非ウイルスベクターを大きく凌駕した。その上、正電荷リポソームにバイオ・サーファクタントを含有させると遺伝子導入効率が飛躍的に増大することを発見した。そこで、このバイオ・サーファクタント含有リポソームの遺伝子治療に応用するための基礎データの蓄積、遺伝子導入機構の解明、ならびに、動物を用いた遺伝子治療の実験を行った。その結果、(1)バイオ・サーファクタントの分子構造(分子種)により遺伝子導入の効率が大きく増大することを発見した。(2)中でも、バイオ・サーファクタントMEL-Aが最も高い遺伝子導入効率を与えた。(3)共焦点レーザ顕微鏡を用いた実験から、MEL-Aはカプセル化した正電荷リポソーム・DNA複合体の細胞膜への吸着、細胞内への取り込み過程を顕著に促進した。(4)担がんマウスの実験から、バイオ・サーファクタント含有正電荷リポソームは遺伝子治療の研究に大きく寄与すると考えられた。
