研究概要

アデノ随伴ウイルス(AAV : adeno-associated virus)ベクターは、安全性が高く、骨格筋などの非分裂細胞へ遺伝子導入可能で、長期的な遺伝子発現が期待できる。そこで、抗腫瘍血管療法や癌の転移・播種の抑制を狙った治療法への応用が期待され、本研究では以下の研究を行った。1)AAVベクターの血清型と組織特異性に関する検討:筋注法に用いるAAVベクターの最適な構築を検討した結果、1〜5型の血清型の中で1型が最も効率よく、またCMVプロモーターを用いることにより筋肉で良好な遺伝子発現が得られることを確認した。経時的観察では、長期間に亘る遺伝子発現が期待できることが判明した。2)バキュロウイルスを利用したAAVベクター作製法の開発:AAVベクターの普及を阻んできたベクター作製技術の壁を打開するため、バキュロウイルスと昆虫細胞を利用したAVVベクター大量作製システムの開発を行った。本法で作製したAAV1-GFP、AAV5-GFPは、従来法で作製したものとほぼ同機能を有していることが遺伝子導入実験から示唆された3)AAVベクターを用いた癌に対する遺伝子治療ストラテジーの検討:応用研究では、卵巣癌の腹膜播種と癌性腹水の制御を目的に、NK4 (HGFアンタゴニスト)、可溶型Flt-1、IL-10 (interleukin-10)の各遺伝子を搭載したAAVベクターを作製し、担癌ヌードマウスを用いた遺伝子治療モデル実験を行った。その結果、特に後2者の有効性を明らかにすることができた。尚、この実験では、卵巣癌細胞の皮下接種あるいは腹腔内接種の前に、AAVベクターの筋肉内注射を行った。これは、手術的に癌病巣を摘出し、遺伝子治療で転移や再発を抑えるという治療法を想定したものである。
