研究概要	1)細胞周期の進行にともなう液胞構造の再構築過程の解析液胞は高等植物の多くの細胞で発達して大きな体積を占めている。このことから液胞は細胞分裂、形態形成、形態制御といった形の変化をともなう現象に深く関与すると考えられる。本研究では液胞膜の観察系として、AtVam3pとGFPの融合タンパク質を恒常的に発現するタバコBY-2細胞の形質転換株BY-GV7を確立するとともに、連続光学切片から立体再構築を行なうプログラムSSRを開発し、細胞周期の進行にともなう液胞の動態を解析した。その結果、M期において分裂装置の周囲にチューブ状の液胞膜の構造TVMが出現し、このTVMがM期の二分された巨大液胞をつなぎ、その内腔間を連絡していることなどが明らかになった。 2)シロイヌナズナ培養細胞を用いた管状要素分化誘導系の確立と微小管の機能の解析管状要素の分化では、二次細胞壁が特徴的なパターンで形成される。この二次壁パターンの制御には微小管が関わっていることが知られている。本研究では、二次壁形成時の微小管の動態を経時観察すべく、tubulinとGFPの融合タンパク質を恒常的に発現させたシロイヌナズナの懸濁培養細胞AC-GT13を作成し、管状要素の分化誘導を試みた。また、微小管と二次壁の立体構造解析を行った。オーキシンを除去しブラシノステロイドを添加した培地で培養することで、AC-GT13を分化させる実験系の確立に成功した。この培養系では2〜3日後から管状要素の分化が始まり、4日後には約30%もの細胞が管状要素に分化した。培養後3日目の細胞を観察したところ、微小管の束は当初は細い二次壁の直下に存在していたが、二次壁肥厚にともない、微小管の束は2つに別れて二次壁を挟み込むように二次壁の縁に沿った位置に変化した。二次壁が肥厚を続けている間、2つの微小管束が二次壁の縁に沿うように幅を広げていく様子が観察された。
