骨髄-腸上皮相互作用による腸管上皮幹細胞システム制御機構の解析

研究概要	炎症性腸疾患をはじめとする腸管粘膜上皮に傷害を来す疾患に対し、上皮再生を促進する新規治療法の開発を目指し、腸管上皮細胞の分化、再生過程につき研究を進めてきた。この過程でGVHD、潰瘍などによる組織破壊修復過程において、ヒト骨髄細胞が腸管上皮として定着し、上反再生に関わる可能性を報告した。本年度の研究では、骨髄由来細胞がどのような性質の腸管上皮になり得るかを骨髄移植後のヒト消化管生検検体を用いて更に詳細に解析した。その結果、骨髄由来細胞は腸管上皮幹細胞ではなく増殖帯に存在する前駆細胞として分布し、増殖、分化する経路を辿ることが推察された。この骨髄由来上皮細胞はいずれも単核であり、multi-colour FISHの結果からも細胞融合の結果であることは否定的であった。さらにGVHDからの粘膜修復過程に於ける解析から、骨髄由来の上皮前駆細胞からの増殖は粘膜修復過程において促進されるとともに、通常吸収上皮型に制御されている分化の方向付けが、粘膜修復に伴い分泌型の上皮細胞へも誘導されることが明らかとなった.マウス腸管において、吸収上皮型と分泌型上皮の分化の方向付けの決定にはNotch Signalの下流で働く複数の転写因子群の関与が推察されている。既にヒト腸管上皮においてもNotch1とその下流の複数の転写因子がCrypt内に発現していることを確認しており、骨髄由来上皮細胞の分化過程の初期段階においてもこれらの分子群が関与し得ることが示唆される。今後、骨髄由来細胞が機能をもった上皮細胞として分布するに至るNotch signalをはじめとする分子機構を明らかとすることにょり、腸管上皮傷害に対する上皮再生を標的とした新規治療法の開発につながるものと期待される。
