免疫不全マウスへのGFP標識臍帯血細胞移植による多分化能、クローナリティー解析

研究概要

我々の開発した免疫不全マウスNOD/SCID/γcnullマウスは、移植ヒト臍帯血細胞の生着を許容し、移植後長期にわたって骨髄、末梢血においてヒト造血機構の再構築が達成できることが明らかとなった。さらに、T細胞を含む機能的免疫学的機構も再構築できることも、我々の研究により示された。この結果を踏まえ、NOD/SCID/γcnullマウスを、種々のヒト幹細胞の分化能や可塑性のin vivoでの評価系としての確立することが期待されており、その確立はヒト幹細胞を用いた再生医療への展開において必須の基盤技術と考えられる。今回、このマウスを用いて、移植ヒト臍帯血細胞より、肝臓、骨格筋等への分化の可塑性について検討した。ヒト臍帯血より未熟細胞であるCD34+細胞を分取移植し、移植後、1ヵ月、2ヵ月、3ヵ月時に、骨髄、末梢血、肝臓、骨格筋、心筋、肺、脳等、種々の組織において、組織特異的分化を果たしたヒト細胞の同定を試みた。従来の報告通り、骨髄、末梢血等の造血組織においては、移植ヒト細胞に由来する血球の出現を確認できた。しかし、肝臓、骨格筋、心筋、肺、脳等の組織においては、血液成分としてのヒト細胞は同定できたものの、組織特異的細胞の出現は確認できなかった。組織再生は、組織障害に引き続いて観察される。そのため、選択的肝臓障害を誘導するCCl4投与、抗Fas抗体投与により、ヒト細胞の肝組織への分化の有無について検討した。その結果、いずれの障害においても、移植ヒト臍帯血細胞に由来する肝細胞の出現を確認した、今後、分化の細胞学的、分子学的機構、機能的評価について、検討を進めていく。
