研究概要	生体は、免疫監視情報を獲得し、それに基づいて免疫応答の方向と様式を決定する。そのための受容体とその制御系を解析した。TCRを介する抗原認識シグナルと共に接着分子LFA-1を介したシグナルが免疫シナプスを形成し、活性化のco-stimulationを導入する。LFA-1を介するoutside-inシグナルはADAP/JNKを介してアクチン再構成を誘導し、免疫シナプス・活性化に不可欠なことを示した(斉藤)。抗原認識の新しい受容体として注目されるペア型レセプターとして新たにPILR等を、さらに、PILRのリガンドとしてPILR-Lをクローニングした。PILR-Lは、IL-4産生細胞株やTh2に高い発現が認められた。PILR-L発現細胞によってDG等を活性化されることがわかり、ペア型レセプターPILRは、T細胞制御に重要な機能を担っている可能性が考えられる(荒瀬)。抗原特異的B細胞が末梢リンパ組織で抗原の刺激を受けて活性化し、成熟していく過程で、胚中心B細胞が抗原に対して特異的な高親和性抗体産生をする。この機能に胚中心関連分子GANPが必要であることを明らかにしてきたが、この際のB細胞におけるGANPの機能を解析した結果、より親和性の高いBCRを発現した抗原特異的B細胞の活性化ではより強力なDNA損傷を引き起こすこと、そのようなB細胞の生存にはGANPが必要であることを明らかにした(阪口)。免疫監視機構の中心的な役割を果たすT細胞の反応性は、抗原提示細胞およびサイトカインによって調節される。サイトカイン(特にIL-6)によるマクロファージと樹状細胞DCの成熟・活性化の制御を検討した。マクロファージの成熟活性化はIL-6信号にて影響を受けなかったが、DCではIL-6信号によって成熟・活性化が抑制されることが判明し、IL-6による制御が、マクロファージとDCで異なうことを示した(村上)。
