生殖細胞における性決定と雄性生殖幹細胞の増殖・分化メカニズム

研究概要

我々はX染色体上にGFP遺伝子を組み込み全身の細胞が緑色の蛍光を発するマウスを用いて受精後2日以内の胚で性の分別を可能にする系を確立している。その系を用いて雌雄のキメラマウスを作製すると、雌雄の生殖細胞を分別できるキメラマウスができる。このようなマウスの精細管内の雌胚に内来する精原細胞様の細胞の性状を詳しく検討した。この細胞は明らかに雄型のゲノミックインプリンティングをうけており、XXという核型にもかかわらず雄としての分化を遂げていることが分子生物学的に明らかになった。また、精巣内に現れる巨大な雌性細胞を検討したところ、卵子に特有の構造である透明帯をもつことが明らかになり、さらに精子との融合能も有していることが判明した。この細胞のゲノミックインプリンティングは不完全ながら雌型であり、同じ精巣内において、雄型および雌型のふた通りの相反するゲノミックインプリンティングがおこることを明らかにした。
In vivo electropration法により精巣支持細胞に増殖因子GDNFを導入し、精巣生殖幹細胞を増殖させ、特異的なマーカーを利用し、FACSにて効率的に単離する方法を確立した。この系を利用し、生殖幹細胞を構成する2つの細胞群(Oct-3/4(+)細胞とOct-3/4(-)細胞)が、その相互転換能により同等の幹細胞を示すことを明らかにした。さらに、このOct-3/4(-)細胞が、未分化と分化の中間の状態にあり、組織再生時にGDNFにより選択的に増殖することを明らかとした。
