研究概要

始原生殖細胞発生全過程、全能性胚細胞における遺伝子発現研究のため、これらの細胞特異的にレポーターを発現するマウスを用いて、始原生殖細胞発生のほぼすべての段階からPGCを単離し、それを材料に16種類のcDNAライブラリーを作製した.これまで、6種の発生段階のライブラリーから計8万クローンを単離し、18000余のEST配列を取得し,その分類を行った.PGC cDNAライブラリー中には新規遺伝子、PGC特異的発現を示す遺伝子が多数含まれることが明らかとなったが,既知遺伝子ではあるが,これまでPGCにおける発現が知られていない遺伝子も多数存在した.例えばその中の一つ,ERR-β(Estrogen related receptor-beta)はこれまで,胚体外外胚葉の一部の細胞で特異的に発現するが,他の細胞,組織では発現していないと考えられていた.しかし,ERR-βはPGCライブラリーにて複数回,ESTが発見され,その後の発現解析の結果,生殖隆起に到達後の始原生殖細胞において特異的に発現していることが確認された.ノックアウトマウスを用いた解析から,ERR-βはPGCの増殖に関与する,との示唆を得た.このように,PGC cDNAライブラリーとそこから得られる遺伝子情報は始原生殖細胞解析のための恒久的研究資源として非常に有用なものと考えられる.現在、これらのEST配列を元にPGCに発現する遺伝子プローブを搭載したcDNAマイクロアレイを作製し,網羅的な遺伝子発現解析を行っている.これまで,ES細胞と始原生殖細胞では共通した遺伝子が発現していると考えられていたが,発現レベルを考慮すると,やはりPGCはES細胞とは異なる発現プロファイルを持つことがわかってきた.
