ミクログリアによるグルタミン酸を介した神経伝達の動的制御

研究概要	今年度は、ミクログリア培養上清のグルタミン酸を介した神経伝達に対する作用を電気生理学的に解析した。まず、新生仔ラット(3日齢)の大脳皮質より調整した初代培養ミクログリアを無血清培地において2日間の培養を行い、培養上清(MCM)を回収し1日間の透析を行った。次に成熟ラットの大脳皮質スライス標本(厚さ400μm)を用い、MCMによるNMDA受容体を介した神経伝達に対する作用をホールセルモードのブラインドスライスパッチ法により解析した。膜電流固定下に大脳皮質第2〜3層ニューロンにおいて白質刺激により誘発される興奮性後シナプス電位(EPSPs)に対する影響を調べた結果、MCM(10%)の適用によりNMDA受容体を介したEPSPsの増大が認められた。次に、膜電位固定下に様々な膜電位におけるNMDA受容体を介した興奮性後シナプス電流(EPSCs)を計測し、電流-電圧曲線を作成した結果、MCM(10%)は測定した全ての膜電位においてNMDA受容体を介したEPSCsを有意に増大することが明かとなった。さらに、幼若ラットの大脳皮質より急性単離したニューロンを用いてホールセルモードのパッチクランプを行い、Yチューブ法を用いて適用したNMDAあるいはグルタミン酸によって惹起される内向き電流に対するMCMの影響を検討した。その結果、MCM(10%)はNMDAならびにグルタミン酸によって惹起される内向き電流をいずれも有意に増大させたが、グルタミン酸に対するMCMの増強程度はNMDAに対するものよりも大きかった。興味深いことにMCM存在下ではNMDAならびにグルタミン酸はいずれも通常では観察されない一過性の大きな内向き電流を惹起した。以上の結果より、MCMはNMDA受容体を介した反応を直接的に増強することが明かとなった。また、その増強作用はMg阻害の緩和によるものではなく、NMDA受容体のグルタミン酸に対する親和性を増大せることによることが強く示唆された。
