研究概要

シナプス後肥厚部(PSD)に存在する多様な分子群の動態を網羅的に解析する為に、まずPSDに存在する分子群をその分子構造と機能から7つのグループに分け、それぞれのグループを代表する分子について、GFP融合蛋白質の作成およびPSD動態に与える影響の解析を試みた。(1)グルタミン酸受容体分子に関しては、AMPA受容体サブユニットGluR1分子、NMDA受容体サブユニットNR2A, NR2B分子、代謝型グルタミン酸受容体サブユニットmGluR5分子のGFP融合分子を作成し、組み換えadenovirusを作成した。(2)細胞接着分子に関してはN-cadherinおよびCNRのGFP融合蛋白質および組み換えadenovirusを作成した。(3)受容体に直接結合する足場蛋白質に関してはPSD-95,PSD-Zip45のGFP融合分子および組み換えadenovirusを作成した。(4)受容体と間接的に結合する足場蛋白質に関してはGKAP, Cortactin-binding proteinのGFP融合分子および組み換えadenovirusを作成した。(5)受容体型リン酸化酵素についてはephrinのGFP融合蛋白質の作成および機能解析を行った。ErbB4について機能変異体を作成、本リン酸化酵素によるPSD分子の動態の制御機構を解析中である。(6)細胞質内リン酸化酵素についてはCaMKIIおよびfyn分子について、これらの分子によるPSD蛋白質の動態制御機構を解析中である。(7)細胞骨格蛋白質についてはactinおよびcortactinのGFP融合蛋白質および組み換えadenovirusを作成した。以上の7つのグループの内、実際の動態解析についてはこれまでに(2)(3)および(7)のグループについての動態の基礎データが得られた。
