研究概要	カルシニューリン(CaN)はカルシウム/カルモジュリンにより活性化される蛋白質脱リン酸化酵素である。そのターゲットとしては細胞質にあるNFATなどが知られており、それらのリン酸をはずすことによって、CaNとコンプレックスとなって核に移行し、MEF2やAP-1と一緒に転写活性の調節を行う役割を担う。これらの転写制御因子の下流にあるのが筋の遅筋化に関する蛋白質であり、ミオグロビンや遅筋タイプの収縮調節蛋白質の発現レベルを上げることが知られている。今回、中程度の運動がカルシニューリンカスケードに与える影響を調べるため、ヒトを対象として最大酸素摂取量の50%に相当する運動強度で1時間の自転車エルゴメーターを使った運動を行わせた。運動前と運動後4時間目の外側広筋バイオプシーサンプルからRNAを採取し、RT-PCR法により以下の分子のmRNAの発現レベルの解析を行った。その結果、c-fosはどの被験者でも上昇が見られたが、NFAT1-NFAT3はそれぞれの被験者でまちまちの反応性を示した。一方CaNやMEF2の発現レベルは変わらなかった。このことにより中程度の運動1時間でもCaNカスケードを惹起することが判明した。そのほか、高地で暮らしているシェルパなどの適応現象にはHypoxia-Inducible factor 1-alfa等が関与していることも分かった。また、低圧・低酸素下においてスーパーオキサイドアニオンを生じせしめる好中球の増加が観察された。さらに、筋の組織切片において、簡単に有酸素能力の高い遅筋線維を見分ける方法も見いだした。以上、運動による骨格筋の筋線維分化・筋代謝能変化に影響を及ぼす分子に関わる知見をもとにさらに研究を進めていく予定である。
