研究概要

1)リンゴ単為結実品種は花器官変異を持っており、花器官形成遺伝子MdPIの発現が欠損していた。しかし本研究で、このMdPI遺伝子の発現組織、発現制御を解析した結果、花器官変異の説明は出来るが単為結実とは直接結びつかないことがわかった。MdPI遺伝子の機能は他の植物のPIホモログのものとよく似ており進化的な保存性が高いことも分かった。
2)MdPI遺伝子が転写調節遺伝子のMADS遺伝子ファミリーに属しているためMdPIの発現抑制により発現に影響を受ける遺伝子を探索した。同じ花器官形成遺伝子のホモログMdTM6、MdMADS13のクローニングに成功した。またこのMdMADS13の発現が単為結実品種で減少し、かつ発現組織も結実と関与する子房や胚珠で観察され、単為結実と関与することを示唆した。
3)正常品種のMdPI、MdMADS13など単為結実に関与する遺伝子の機能を調べるために、これらの遺伝子発現の抑制、または促進した組み換え体リンゴの作出を行った。これまでのリンゴの形質転換効率は非常に低いものであったが、本研究では形質転換法や転換体の培養法、形質転換に適した品種の選抜を行うことで、これまでより数十倍高い形質転換効率を持つ系を確立することに成功した。
4)MdPIのアンチセンス、またはサイレンシングベクターを組込んだ組換えリンゴの作出に成功した。またMdMADS13のアンチセンスベクターを導入した組換え体リンゴ、さらにMdTM6のアンチセンスベクターを導入した組換えリンゴの作出に成功した。
5)これら組換えリンゴが開花、結実して初めて導入遺伝子の機能が解析できるため、早期開花組換えリンゴを作出し、2ないし3年で開花することに成功した。
