大腸菌におけるリポ蛋白質の選別輸送と膜局在化の分子機構

研究概要	大腸菌は、内膜または外膜のペリプラズム側にアンカーした90種以上のリポ蛋白質を持っている。+2位にAspを持つ内膜特異的リポ蛋白質が、どうして内膜にとどまるかは明らかでなかった。内膜特異的リポ蛋白質を外膜に運ぶ変異株を分離し、ABCトランスポーターLolCDE複合体のサブユニットLolCに変異があることを明らかにした。リポ蛋白質の+2位を化学修飾し、LolCDEと共にプロテオリポソームに再構成し、リポ蛋白質特異的分子シャペロンLolAとATPに依存した遊離反応を解析した。また、再構成に用いるリン脂質を化学修飾した。これらの解析によって、Aspは正電荷を持ったリン脂質であるホスファチジルエタノールアミン(PE)と静電的相互作用をし、N-末端の脂質修飾されたCysとPEが水素結合するため、強固な複合体が形成され、これがLolCDEによって遊離されない原因であることを明らかにした。ATPaseサブユニットLolDと膜サブユニットLolE、LolC間にはリポ蛋白質の遊離に重要な相互作用が推定されていたが、変異体の解析によって相互作用領域を明らかにした。LolAとリポ蛋白質受容体LolBは、よく似た疎水性キャビティーを持っているが、LolAの疎水性キャビティーには蓋があることを結晶構造によって明らかにした。さらに、蓋の開閉はリポ蛋白質の結合と蓋を持たないLolBへの受け渡しに重要であることを、LolA変異体の解析によって明らかにした。外膜に局在するリポ蛋白質LolBの外膜へのアンカーはLolBの機能に必須ではないこと、LolBの蛋白質部分には、脂質と相互作用しリポ蛋白質を膜に組み込む機能があることを明らかにした。
