医用フェライト・ナノビーズの診断・治療・分析への応用展開

研究概要

マウスに免疫して、ハイブリドーマ由来のマウスIgGIクラスの抗CEAモノクローナル抗体を作製した。このCEA抗体を直接フェライトビーズに固定すると、その構造が変換し、抗原との結合能が失われる可能性があるので、我々はIgG抗体と選択的に結合するプロテインGをまずフェライトビーズに固定化することを考えた。そこで、プロテインGを添加したFeCl_2反応溶液を用い、Fe^<2+>→Fe^<3+>酸化反応を利用して合成反応を行った。得られたフェライト・ナノビーズの表面にプロテインGが固定されていることが明らかとされ、またゼータ電位・粒子計測装置で測定したところ、プロテイン固定化ナノビーズは良好に水中に単分離していることがわかった。さらに、フェライトビーズに固定化されたプロテインGは坑CEA抗体と効率よく結合し、結合した坑CEA抗体は抗原との結合能を保持していることを確認できた。また、約30塩基長の一本鎖DNAをその末端を介してフェライトビーズ表面に固定化する技術を開発した。それを用いて、固定化した一本鎖DNAに相補性のDNA断片をハイブリッドさせる技術を開発した。
以上によって得られた抗体固定ナノビーズを用いたハイパーサーミア実験の準備およびDNA単鎖固定ナノビーズをホール効果で検出する準備を進めている。
