研究概要	今年度は、計画したACTH、レプチン、LIF/gp130のそれぞれの系について、下記の通り実験を進めた。 1.ACTHの胎生期における作用についてまず、ACTH受容体の胎生期における局在を免疫組織化学的に解析した。中枢神経系、分岐型実質臓器を含む多くの臓器に、受容体分子種により機能的な分化を示唆する特異的な分布パターンを観察した。ACTH産生腫瘍細胞株AtT-20細胞のマウス胎児への注入実験を進めたところ、注入個体の肺、腎臓において上記の受容体発現パターンに基本的に対応した部位に、ACTHの直接・間接的効果を示唆する予備的な所見を認めた。 2.レプチンの胎生期における作用についてまず、レプチン欠損ob/obマウスと野生型マウス胎児の大脳における神経細胞数を計測し、神経幹細胞や前駆細胞への一定の効果が示唆された。これを確認するため、大脳由来の神経幹細胞の培養系において、レプチン投与の効果についてさらに検討を進めている。 3.LIF/gp130の胎生期における作用について gp130の胎生期中枢神経系におけるリガンドとしてのLIF、および同様に胎生期脳の神経細胞に対して増殖刺激を与える因子であるFGF2について、その発現ならびに産生部位を免疫組織化学的に検索した。また脳脊髄液中におけるこれらの分子の存在につき、ウェスタンブロッティングおよびELISAにて解析し、その存在と時期による消長のパターンを観察した。したがって、脳脊髄液を介した脳神経上皮細胞の増殖調節の可能性が示唆された。
