マウス嗅球の新生介在神経細胞による神経回路形成の分子機構の解析

研究概要

我々が樹立したNestin-GFPマウス嗅球には2種類のGFP陽性神経細胞が存在する。GFP発現の弱い細胞をtype W細胞、強い細胞をtype S細胞と呼んでいる。Type W細胞は、嗅球内を移動中の未分化穎粒細胞である。type S細胞は、細胞分裂後1か月以上経過しているが、典型的な成熟顆粒細胞とは異なる形態をもつ顆粒細胞である。
1.GFP陽性顆粒細胞の発生過程の解析
マウス新生仔のType W, type S細胞の発生過程を検討した。生後7日ではGFP陽性細胞は全てType W細胞であった。生後14日では殆どがType W細胞で、ごく少数のtype S細胞が観察された。生後21日では多くのtype S細胞が存在した。Type W細胞の割合は、出生直後が最も多く、経時的に減少した。
2.type W細胞の解析
未分化神経細胞が脳実質内を移動する分子機構を解析するため、type W細胞を嗅球培養スライスで観察する方法、cell sorterで回収する方法を確立したい。発生過程の解析より、生直後から7日までの嗅球を用いるのが望ましいことが分かった。現在、スライス培養、sorterによる細胞回収の方法を検討中である。
3.type S細胞の解析
type S細胞の形態を詳細に観察するため、固定嗅球スライスを用いて、GFP蛍光を指標に、type S細胞に更に蛍光色素をガラスピペットで注入する系を確立した。この結果、type S細胞が多くのスパインを形成していること、またその樹状突起の先端が、嗅球の投射ニューロンである僧帽細胞の細胞体に接着していることが分かった。これは通常の顆粒細胞が僧帽細胞の樹状突起とシナプス形成している点と大きく異なっていた。これらから、type S細胞は新規のタイプの成熟した顆細胞であることが判明し、嗅球情報処理において特別な機能を担っていると考えられた。
