研究概要	本研究において、リン酸ERKは正常のニューロンではほとんど発現していないが、ラット足底にカプサイシン注射2分後に、多数のDRGの小型細胞でERKのリン酸化反応が観察される。この全く新しいシグナルのキナーゼ経路を解明することが一つの目的である。また、ERKリン酸化を用いたTRPファミリー分子やATP受容体の一次感覚ニューロンにおけるin vivo機能解析を目指して、末梢からの種々の(熱、冷、機械的、化学的)侵害刺激に対するERKのリン酸化反応が、どのような疼痛関連分子を持つニューロンで生じるかを決定する。研究実績の概要は以下の通り。・P2X/P2Y受容体の各サブタイプのcDNAをクローニングした。・cRNA probeによるin situハイブリダイゼーション法を用い、DRGにおける詳細な発現を確認した。タイプ別に特異的な発現パターンを示すことを明らかにできた。・末梢への各種侵害刺激(熱、冷、機械的、化学的)後の、DRGにおけるERKリン酸化反応の免疫組織化学法と、P2×3抗体との二重染色を施行した。その結果、末梢炎症組織に対する機械的刺激において、P2×3発現ニューロンにおけるERKの発現が、劇的に増加することが明らかとなった。また、P2×3アンタゴニスト投与で、機械的刺激に対するERKの発現が減少した。・VR1受容体cDNAより、in vitroトランスクリプションキットを利用してVR1 mRNAを作成し、このmRNAをOocyteにマイクロインジェクションし、VR1蛋白を強制発現させた。・2日後から2週間の間に膜電位固定法、パッチクランプ法により電気生理学的測定が可能となった。測定は、(1)カプサイシンによる電流の測定、(2)繰り返しのカプサイシン刺激による電流のPotentiation、(3)MEK Inhibitorを投与しPotentiationの変化を現在観察中である。
