日本人2型糖尿病感受性領域の連鎖不平衡マッピングによる糖尿病感受性遺伝子の同定

研究概要	日本人の罹患同胞対を利用した全ゲノムスキャンにより同定した計9箇所の染色体領域(1p36-P32,2q34,3q26-q28,6p23,7p22-P21,9p,11p13-P12、15q13-q21と20q12-q13)の中で、アディポネクチン遺伝子が感受性遺伝子であることを既に明らかにした3q26-q28以外の領域に含まれる候補遺伝子についてSNP(single nucleotide polymorphism)による相関解析を行った。1p36-p32領域にはインスリン感受性ホルモンであるアディポネクチンの作用を媒介するAMPキナーゼ遺伝子が存在し、α1、α2両アイソフォームのSNPを保持することによりインスリン抵抗性が有意に上昇することを見出した(Diabetologia, submitted)。また最近単離・同定に成功したアディポネクチン受容体遺伝子1,2(AdipoR1,AdipoR2)について最低10kbに一個の割合でSNPを選択し詳細な相関検討を行っている.11p13-p12領域についてはpaired box gene 6(PAX6)、15q13-q21領域についてはカルパイン3(CAPN3)についてSNPによる相関解析を行ったが日本人においては2型糖尿病の主要な感受性遺伝子とは認められなかった。20q12-q13領域のhepatocyte nuclear factor 4α遺伝子(HNF4A)については2型糖尿病と有意に相関するSNPを同定し、現在つの独立したDNAパネルで再現性を検討中である。
