細胞情報伝達阻害物質の探索と天然分子プローブの創製

研究概要	未解明の細胞内情報伝達機構を詳細に明らかにすることが、細胞内情報伝達機構の破綻による疾病の発症メカニズムを解明し、新たな治療法の開発につながると期待される。本研究では、特にがんと神経変性疾患に着目し、これら疾患に関わる細胞内情報伝達系に特異的に作用する機能性低分子化合物を探索するアッセイ系を構築し、これまでに集めた海綿を中心とする海洋生物の抽出エキスライブラリーから探索する。そして新たに発見した阻害物質とそれらから設計・合成した光親和性放射性標識体やビオチン標識体などの各種の天然分子プローブを用いて標的分子を同定し、細胞内情報伝達系への作用機構を解析する。さらに、見出した微量活性物質については、細胞情報伝達系解析のための新しいツールとして広く世に供給するために全合成研究を展開する。今年度は、すでに、Neuro 2A細胞に対する分化誘導物質として発見している海綿由来のポリケチドlembehyne Aについて、構造活性相関の知見をもとに標的分子を同定するために、分子末端に光反応性のアジド基と放射性同位体でラベルした分子プローブを設計合成した。そして、Neuro 2A細胞に特異的に結合する蛋白の検出に成功した。また、ヒト慢性骨髄性白血病細胞(K562)を赤芽球様細胞へと分化誘導する物質として、海綿からセスキテルペンキノンsmenospongine類を見出し、作用の詳細な解析や、構造と活性の相関を解析した。また、先に構築したP-gpやMRP1に代表されるABCトランスポーターを過剰発現する多剤耐性がん細胞(KB-C2,KB-CV60)を用いるアッセイ系を用いて、海綿から多剤耐性克服活性を有する脂溶性ペプチドを見出し、その平面構造を決定した。
