光合成アンテナをモデルとした超分子ナノデバイスの創製

研究概要

光合成細菌の膜外アンテナ部におけるクロロフィル自己集積体のモデルとして、多様な新規クロロフィル誘導体を合成し、その自己集積体を調製した。その超分子構造を、より詳細に解明するために、情報量の多い核磁気共鳴法を用いることにした。カドウミウニム錯体1は、固体状態で秩序正しい自己会合体を形成することが知られている。そこで、全ての構成炭素と窒素原子を^<13>Cと^<15>Nで同位体標識したクロロフィル-aを原料として、全^<13>C^/^<15>Nを全て標識したU-^<13>C/^<15>N-1を調製した。この固体状態は、無標識体と同様に自己会合体を形成することが、可視吸収スペクトルから判明した。この固体の三次元多核磁気共鳴スペクトル(^<13>C-^<13>C-^1H)を測定することで、分子間の距離情報を求めた。このデータは、これまで想定していた「中心カドミニウムに配位結合した3^1位の水酸基が、13位のケトカルボニル基に、水素結合しつつ、クロリン環同士がππ相互作用する」ことから見積られた超分子構造を、強く支持するものであった。
次に、クロロフィル骨格で重要な役割を果たしている酸素を含んだ官能基の情報を得るために、^<18>Oで標識されたモデル化合物を、位置選択的かつ高効率的に合成することにした。ホルミル基やケト基のようなカルボニル基を、アセタールやケタール保護した後、酸性の^<18>O水で処理すると、高効率で^<18>Oがカルボニル基に導入できた。また、カルボニル基を直接酸性の^<18>O水で処理しても、同様の^<18>O標識化合物が合成できるが、標識率はやや落ちた。但し、後者の手法の方が簡便であり、その適用範囲も広いという利点があった。^<18>Oで標識されたカルボニル基を、振動分光法(赤外吸収/共鳴ラマン分光法)で検討したところ、無標識のものに比べて、約30cm^<-1>低波数側にピークが移動することが判った。
