ミトコンドリアの母性遺伝:雄性mtDNAの選択的分解機構の可視化とその分子機構

研究概要	申請者らは、同形配偶子生殖の真正粘菌Physarum polycephalumを用いて、接合子内で片親由来のミトコンドリア核(mt核,DNAタンパク質複合体)が選択的に分解され、mt核を消失した片親由来のミトコンドリアは、30時間以上を経過してようやく分解されることを明らかにした。ミトコンドリアの母性遺伝機構を解明するためには、1)mt核の選択的消失に直接関るヌクレースを単離し、2)片親のミトコンドリアあるいはmtDNAに対するその選択性がいかにして発揮されるのかを調査すべきであろう。ミトコンドリアの崩壊が先だとすれば、母性遺伝の選択性は、雄性ミトコンドリア崩壊を選択的に誘起する酵素群にあることになる。その意味で、mt核の消失が先か,ミトコンドリアの崩壊が先かは極めて重要な問題であり、多くの生物を用いて詳細に検討する必要があろう。 15年度は、mt核が容易に観察できる真正粘菌に加え、緑藻網からヒラアオノリEnteromorpha compressaと高等植物としてヒロハノマンテマSilene latifoliaを用いて、原始的な同型配偶子生殖から、異型配偶子生殖を経て、花粉生殖の高等植物へ至る進化過程で、mt核消失とミトコンドリア崩壊の時期を特定し、ミトコンドリアの遺伝様式ともに雄性ミトコンドリアの排除機構の変遷を分子細胞形態学的で解析した。1)従来用いられていたmtDNAのRFLPの他に定量PCR法によってミトコンドリアの遺伝様式を詳細に再検討した。また、2)配偶子形成と接合子過程における雌雄ミトコンドリアの挙動、特に、mt核の選択的消失とミトコンドリア崩壊の時期的関係を、蛍光抗体染色法などを用いて詳細に観察した。また、3)真正粘菌でmt核の選択消失に直接関ると予測されるヌクレースとその選択性をアッセーする雌雄の単離ミトコンドリアやmt核あるいはmtDNAを用いたシステムを構築した。15年度は、真正粘菌のミトコンドリアの片親遺伝を誘発するMn^<2+>要求性のDNaseの単離精製を最優先し、mt核消失とDNase活性、DNaseの選択性、出現時期、細胞内分布などを中心に調べた。
