研究概要	大腸菌RNAポリメラーゼは、7種類のシグマ因子、300種類の転写因子との相互作用で、遺伝子認識特異性が変り、その結果、ゲノムの発現パターンが変化する。転写包括制御の全体像の解明を目的とした研究を実施し、本年度次の成果を得た。研究は、順調に進展し、原核生物の転写研究のブレークスルーに成功した。 1)シグマ因子、転写因子遺伝子欠損株のプロテオーム、トランスクリプトープ解析から、各因子の支配下の遺伝子群を推定した。 2)全シグマ因子、約150種類の転写因子を発現、精製した。純化転写因子をゲノムDNA断編と混合して、転写因子に結合するDNA断片コレクションを構築する実験系を確立した。幾つかの転写因子については、DNA配列の分析から、ゲノム上の転写因子の結語部位のマッピングを行った。 3)これら多面的なアプローチで同定した各転写因子の結合サイトを含むプロモーターを単離し、我々が開発した、蛍光タンパク2種類をもつプロモーター活性測定ベクターに挿入し、細胞内での転写因子の影響を解析した。 4)転写因子による調節様式は、再構成RNAポリメラーゼによる単離プロモーター試験管内転写系に、純化転写因子を加えることで解析した。 5)転写過程での転写因子の作用様式は、RNAポリメラーゼとの接点と相関する。先に我々が開発した人工プロテアーゼFeBABEを結合した転写因子と再構成RNAポリメラーゼを利用した転写反応中に切断されたRNAポリメラーゼサブユニットを分析することで決定する実験系を開発した。この系を利用して幾つかの転写因子に関して新に接点を同定した。 6)純化転写因子で家兎を免疫し抗体を作製した。抗体を利用したウエスタン法で、約50種類の転写因子に関して、細胞内濃度を測定し、また培養条件と、培養時間による変動を実測した。 7)二成分系の転写因子に関しては、30種類の純化センサーキナーゼと34種類の純化転写因子を用いたリン酸化反応を解析し、センサーキナーゼと基質転写因子の全ての組み合わせを同定したが、約20の異種二成分系間の組み合わせでも、リン酸化のクロストークがあることを発見した。
