ブロモウイルスの増殖に関与するシロイヌナズナ遺伝子のクローニングと機能解析

研究概要

1.ブロモウイルスの一種であるSpring beauty latent virus (SBLV)はシロイヌナズナCol-0の突然変異体2433B中で細胞間移行が遅延し、この遅延は劣性の形質によって支配されていた。これまでに、原因遺伝子座のファインマッピングに向け、2433Bと他のアクセッション(Nd-1)とのF2種子を取得した。2.SBLV増殖の遅延するシロイヌナズナアクセッション(Pla-0)中の原因遺伝子座は複数存在することが示唆されたため、現在、Pla-0とCol-0との交配後、ライン分けを行って、原因遺伝子のマッピングに向けた準備を行っている。3.SBLV感染により激しい病徴を示すS96と無病徴感染を示すCol-0における病徴発現の違いは不完全優性の核内単一遺伝子座によって支配されていることがわかり、その遺伝子座をSSB1と命名した。SSB1遺伝子座は第4染色体上の遺伝子マーカーCAT2とnga1107の間にマッピングされた。現在,さらにSSB1のファインマッピングを行っている。4.野生型Col-0と変異体cpr5におけるBrome mosaic virus (BMV)とSBLVの感染性を調査した結果、BMVの感染が野生型Col-0中の少なくとも一細胞での増殖のステップで阻害されていることが示唆された。また、CPR5遺伝子に欠損を持つ他の既報の変異体におけるBMV、SBLVの感染性を調査し、これまでの結果がCpr5遺伝子の変異のみによって起こっていることを確認した。cpr5変異体において恒常的に発現している防御応答関連遺伝子の機能をさらに欠失させたdoubleまたはtriple変異体を用いた解析から、BMV感染に対すcpr5変異体の感受性には既知の防御応答経路とは異なる機構が関与していることが強く示唆された。
