ユビキチンシステムによる核内レセプター不活性化機構の解析

研究概要	核内レセプターは、脂溶性ビタミン・ステロイドホルモン・脂質・コレステロール代謝物などをリガンドとするリガンド誘導性転写因子である。核内レセプターは、構造上、機能上の相同性から遺伝子スーパーファミリーを形成していることが知らており、ヒトで50種類弱存在する。近年、これらの核内レセプターがユビキチン・プロテアソーム系で分解を受けることが示された。しかしながら、その分子機構、生物学的重要性については未だに不明である。わたしたちは、リガンド誘導性転写因子である核内レセプター、特にエストロゲンレセプターを材料とし、転写因子のユビキチン化と転写不活性化機構の関連について研究を進めている。エストロゲンレセプター(ER)は女性ホルモンであるエストロゲンを受容し、DNAの特異的配列に結合して、下流に存在する標的遺伝子群の転写を制御する。わたしたちはERがエストロゲン存在下、非存在下においてそれぞれ異なったユビキチンリガーゼによってユビキチン化を受け、分解されることを見出した。エストロゲンの結合していないERはHsc70を含むシャペロン複合体によってCHIPと呼ばれるユビキチンリガーゼと結合し、分解される。CHIPは変異型のERを選択的にユビキチン化することから、ERの品質管理に関与しているものと考えられる。一方、エストロゲンの結合したERは新規ユビキチンリガーゼによってユビキチン化されることが明らかとなった。このユビキチンリガーゼはエストロゲン依存的にERと結合し、ユビキチン化することによって分解へと導く。現在、このユビキチンリガーゼの機能を探るため、遺伝子欠損マウスの作成を進めている。
