胃・腸特異的カルパインの機能解析

研究概要	カルパインは細胞質内に存在し、Ca^<2+>により活性を制御されるプロテアーゼである。細胞骨格系タンパク質、情報伝達系酵素・転写因子などに直接作用し、これらの活性・構造を変換・制御するモジュレータープロテアーゼである。哺乳類細胞では14種類の遺伝子が同定され、スーパーファミリーを形成している。中でも組織に特異的に発現が見られる組織特異的カルパインがいくつか発見されており、その組織との機能と密接な関係が予想されるため、その生理機能に大きな興味が持たれている。我々が発見したnCL-2/-2'やnCL-4は、胃や腸に発現が限定されるのみならず、興味深い構造をもつ選択的スプライス産物が同時に発現している。さらに最近我々は、nCL-2/-2'が胃の上皮層ピット細胞に極めて限定的に局在することを見出し、細胞移動や粘液分泌に重大な機能を果たすことが強く示唆された。胃腸は食品の消毒、免疫、あるいは消化という様々な点で非常に重要な機能を果たす組織であり、それ故に胃酸や粘液の分泌など極めて特殊化した機能を有する。そこで本研究は、遺伝子改変マウスとDNAチップテクノロジーを用いて、胃・腸特異的カルパインの生理機能を解明することを目的とした。まず疾患という生物現象と関連づけるために、遺伝子改変マウスを用いたアプローチを開始した。nCL-2/-2'のC105Sノックインマウスは、共同研究によりホモマウスの作出を完了し、胃・腸の形態学的・機能的解析をおこなった。その結果、ノックインマウスでは、ストレス下での胃出血傾向が、野生型マウスに比べて著しいという興味深い表現型を示すことが判明した。現在、その分子機構を解析中である。
