海洋動物におけるミトコンドリアゲノミクスの展開

研究概要	人為的影響が巨大かつ複雑になる中で、生物多様性を維持しつつ水産生物資源の持続的利用を図る必要性はますます高くなっている。水圏資源生物の集団構造の把握は困難なことが多く、また卵や幼生の種識別はきわめて困難で、これらのことが合理的な資源管理方策を策定する上での妨げとなっている。こうした状況の改善のために、集団や種の信頼の高いDNA標識が強く求められている。本研究は、DNA標識源として重要なミトコンドリア(mt) DNAを取り上げ、これまで実施して来た魚類mtDNA全塩基配列分析研究をさらに推し進めるとともに、研究対象を甲殻類にまで広げ、海洋動物のミトコンドリアゲノミクスの新たな展開を目指すものである。 1年目の今年度は、これまで入手できていなかった魚類および甲殻類の標本収集に努め、約200種の貴重な標本試料を得た。魚類標本の種査定はわれわれ自身でおこない、甲殼類標本の種査定は甲殻類分類学の専門家である千葉中央博物館の駒井智幸氏の協力を得ておこなった。魚類約100種および甲殻類約10種について、当研究グループで確立した効率的なmtDNA全塩基配列決定法を活用して、mtDNA全塩基配列を決定した。甲殻類ではとくに重要な資源生物を含む十脚類(エビ・カニ類)に力点を置いて研究を進めた。得られたミトコンドリアゲノムデータは、すでに集積したデータと照らし合わせながら、その特徴や変異性について検討した。さらに、データ解析のための計算環境を整備した上で、いくつかの分類群に焦点を絞って分子系統解析をおこなった。得られた解析結果に総合的な考察を加え、論文化を進めた。
