マスト細胞のマトリックス接着におけるプロスタグランジンE2の役割に関する研究

研究概要	本研究の目的は、炎症・アレルギーの発症に深く関わるマスト細胞について、どのようなメカニズムで結合組織や粘膜組織中において成熟・分化するかについて解明するために、マスト細胞の前駆細胞モデル細胞としてマウスP-815細胞を用いて、in vitro細胞培養系で同細胞のマトリックスへ定着・遊離メカニズムを検討することにある。本年度は(1)マスト細胞のマトリックスヘのPGE2依存的な細胞接着、と(2)マスト細胞のマトリックス接着による細胞の機能発現に関する研究を行い、以下の研究成果を得た。(1)については、マウスP-815マスト細胞は、フィブロネクチンをコートしたプレートにて培養すると、添加PGE2依存的に細胞接着が誘導されること、この誘導はPGE2受容体のEP4サブタイプによる細胞内cAMPシグナルを介して起こることを明らかにした。このPGE2誘導による細胞接着反応は細胞内Ca^<2+>濃度の持続的な上昇に依存し、細胞内Ca^<2+>の上昇メカニズムはSOC依存的な細胞外Ca^<2+>の流入反応によることがわかった。さらに、細胞内Ca^<2+>の下流はCaMキナーゼによるタンパク質のリン酸化で、Row-MLCK-アクチン系の活性化は関与していないことがわかった。(2)については、マトリックスのフィブロネクチンに接着したマウスP-815マスト細胞は細胞増殖を停止し、マトリックスから遊離すると再び増殖を開始することがわかった。接着している間にどのような機能(サイトカイン生成など)が活性化されているかを検討中である。
