研究概要

Edg5ホモノックアウトマウスは離乳期前後に約半数が死亡する。系統的な全身各臓器の病理組織検索によって、変性を示している臓器を同定した。この臓器障害がホモノックアウトマウスの死亡にどのような意義を有しているかを検討する目的で、この臓器の機能検査を行っている。また、この臓器の機能に影響を及ぼす別の臓器の異常の有無を解析している。これまでの解剖例から、直接死因として血管障害が関与している可能性が考えられる。そこで、血小板凝集反応、出血時間を解析した。現在、観察マウス数を増やしている所であるが、現時点では野生マウスとの間に明らかな差は見られていない。Edg5ホモノックアウトマウスは血圧低下を示すことから、何等かの循環機能障害が死亡に関わっている可能性もある。不整脈に関しては、麻酔下で心電図記録を行ったが、不整脈は今のところ検出されていない。また、心超音波検査を行ったが今の所、明確な異常所見は得られていない。Edg5トランスジェニックマウスを樹立して、現在その表現型を解析している。
Edg5による細胞運動抑制制御に関与するRhoエフェクターの同定を試みた。様々な遺伝子導入実験によって、候補エフェクター分子を絞りこんだ。さらに、候補エフェクター分子の活性化体、抑制型変異体の効果を検討している。また抑制性制御に関与するRac-GAPの同定を目指して、10数種に及ぶRac-GAPの発現を調べ、さらに発現を変化させた場合の機能変化を検討している。
