研究概要

本研究では未分化な網膜細胞に発現している転写因子のプロモーターを単離し、異なる蛍光波長を持つEGFPをその下流で発現させることにより網膜を亜集団に分画し幹細胞を同定しようということを目標とした。平成15年度は転写因子のプロモーターの単離を計画した。当初、BACライブラリーから目的の遺伝子の上流部分の遺伝子のフラグメントの単離をおこなったが充分なプロモーター領域が得られず、BAC-recombinationの系を導入して、BACクローンそのものにEGFPなどを挿入し、トランスジェニックマウスを作製する方針ですでにいくつかの組み替え体を得た。一方で、網膜幹細胞のアッセイ系として、単細胞での定量的な増殖能、分化能などを測定する系の開発をおこなった。それらの系をもちいて、すでに入手したPAX6-EGFPマウスの網膜細胞をセルソーターで分離し増殖能、分化能を検討している。
