研究概要	私共は、接着分子ネクチンと、ネクチンをアクチン細胞骨格に連結するアクチン線維結合蛋白質アファディンから構成され、アドヘレンス・ジャンクション(AJ)に局在する細胞間接着機構を見出している。また私共は、ネクチン-アファディン系がまず細胞間接着を形成し、そこにカドヘリン-カテニン系やクローディン-ZO-1系をリクルートし、AJとタイト・ジャンクションのオーガナイザーとして機能していることを明らかにしている。本年度の本研究において、ネクチン-アファディン系とカドヘリン-カテニン系との相互作用を解析し、以下の成果を得た。 1.細胞外ドメインを欠失させたカドヘリンの変異体(Cadherin-DN)はドミナント・ネガティブ変異体として機能することが知られている。しかし、その作用機構は不明である。私共は、Cadherin-DNを発現している上皮細胞にネクチンを強制発現させると、Cadherin-DNのドミナント・ネガティブ活性が抑制されることを見出した。さらに、ネクチンの強制発現は野生型カドヘリンの細胞接着活性を促進することを明らかにした。したがって、Cadherin-DNは内在性のカドヘリン-カテニン系とネクチン-アファディン系の連結を競合的に阻害して、カドヘリンの接着活性を阻害すると考えられた。同時に、ネクチンはカドヘリンの細胞接着活性を制御していることが示唆された。 2.ネクチンの細胞外ドメインとIgG Fcを融合させたリコンビナント蛋白質(Nefと命名)を作製し、Nefがネクチンのアゴニストならびにアンタゴニストとして機能することを見出した。さらに、このNefを用いて、ネクチンによる細胞間接着がカドヘリンのリクルートに必要であり、かつ十分であることを明らかにした。このように、本年度の本研究は予想以上に進展し、当初の目的をほぼ達成することができた。
