足場依存細胞増殖制御システムの研究

研究概要

癌の本質は、無規律的増殖にある。この特異な増殖性は培養細胞系で、「足場(接着)に依存しない増殖能の獲得」によって再現される。換言すると、正常細胞には、接着を介する高度に組織化された増殖制御機構が存在する事を意味する。今日、増殖因子等による細胞増殖制御機構についてはかなりの詳細が明らかになった。しかし残念な事に、正常細胞と癌細胞の増殖の分水嶺となる「足場依存細胞増殖制御機構」については今だ殆ど明らかでない。我々は、数年来足場依存細胞増殖制御機構に取り組み、独自の解析系を樹立した。この系は、癌遺伝子v-srcで癌化した細胞(SR3Y1)に、細胞膜蛋白質SHPS-1を発現する事により樹立できた。SHPS-1発現SR3Y1は、形態はSR3Y1と変わらず癌化しており、接着状態での増殖も遜色ない。しかし驚く事に、その増殖は強く足場依存となり、ヌードマウスでの造腫瘍性を失っていた。本研究は、これらの実験系を用いて、足場非依存増殖を制御するシグナル伝達系を明らかにしようとするものである。本年は、SR3Y1のフィブロネクチン状への接着の、SHPS-1の有無による変化を解析した。まず、SHPS-1発現をTet inducible systemによって可逆的に制御できるSR3Y1株を樹立した。この細胞を用いて、接着の変化を解析したところ、SHPS-1の発現により劇的に接着が回復する事、この時細胞はruffling構造を回復する事、focal contact様構造が回復する事が明らかになった。現在この制御機構を更に解析中である。
